human TAS2R receptor family

receptor / receptor-tas2r-family

ヒトTAS2R受容体ファミリー

Family of G-protein-coupled receptors involved in human bitter taste detection. Expressed also in tissues outside the tongue (airways, paranasal sinuses, intestines), where they participate in cell-autonomous chemical sensing independent of taste perception (the physiological ligands and detection methods for extraoral function remain disputed).

Reading the bitterness

The human TAS2R receptor family comprises a group of G-protein-coupled receptors that detect bitter compounds in taste buds of the tongue. In experiments expressing mouse and human receptors in heterologous cells, the mouse receptor mT2R-5 responded to cycloheximide, and human hT2R-4 and mouse mT2R-8 responded to denatonium and propylthiouracil. A mouse strain with low sensitivity to bitter compounds showed amino acid substitutions in mT2R-5 that reduced receptor responsiveness. This receptor-level and mouse genetic evidence forms the foundation for understanding that the TAS2R family mediates bitter taste.

It is not uncommon for a single bitter compound to activate multiple TAS2R receptors simultaneously, or for a single receptor to respond to multiple bitter compounds. In receptor assays using cultured cells, caffeine is reported to activate 5 receptors (hTAS2R7, 10, 14, 43, 46), while hop-derived bitter compounds (iso-alpha-acids and prenyl naringenin, among others) activate 3 receptors (TAS2R1, 14, 40). In a bitter fraction from trypsin-digested casein, cells expressing hTAS2R1, 4, 14, 16 responded, with hTAS2R1 and hTAS2R4 showing particularly strong responses. However, this fraction is a mixture of numerous peptides, and individual peptide-receptor correspondences have not been determined.

TAS2R is not exclusively a receptor of the tongue. In experiments using cultured human airway epithelium, motile cilia were found to express T2R4, T2R43, T2R38, T2R46, and addition of bitter compounds elevated intracellular calcium, increasing ciliary beat frequency by approximately 25%. In airway smooth muscle, bitter compounds such as quinine relax muscle far more strongly than the β-agonist isoproterenol; in one experiment, active tension was lost by >90%, whereas isoproterenol resulted in only approximately 30% reduction. However, chloroquine and denatonium used in these experiments required high concentrations (approximately 100 µM) to completely relax pre-contracted muscle, and the authors themselves note the possibility that these are not ideal candidates for clinical application. Although this relaxation was reproduced in another laboratory, explanations conflict between those proposing that local calcium elevation opens potassium channels and those proposing the opposite—that it inhibits those channels and blocks calcium channels—and the mechanism remains unsettled.

What extraoral TAS2Rs originally detect in the body remains, in most cases, unknown. Researchers in this field themselves point out that the majority of bitter compounds used in experiments are not the physiological ligands that these receptors encounter naturally. A rare exception is airway T2R38, which has been shown to respond to quorum-sensing molecules (acyl-homoserine lactones) produced by Pseudomonas aeruginosa and other bacteria, confirmed in human paranasal sinus cultures from genotyped subjects. These molecules are considered to be natural ligands that the paranasal sinus epithelium actually encounters, not food-derived bitter compounds. This single example demonstrates that natural ligands may be found for extraoral TAS2Rs, but this finding does not extend broadly to other tissues.

A review of human bitter taste receptors and their role in perception.
Limitations: An older review; the receptor nomenclature needs to be updated to current usage.

Verbatim: 'we use a heterologous expression system to show that specific T2Rs function as bitter taste receptors.' The mouse T2R (mT2R-5) responds to cycloheximide, and human hT2R-4 and mouse mT2R-8 respond to denatonium and 6-n-propyl-2-thiouracil. A mouse strain with low sensitivity to cycloheximide carries an amino acid substitution in mT2R-5 that lowers receptor responsiveness, and tastant-dependent gustducin activation was demonstrated. Functional and genetic evidence, independent of the Meyerhof group, that TAS2R/T2R receptors mediate bitterness.
Limitations: Receptor-level and mouse-genetics evidence. The link to human perception rests on hT2R-4 responding to PROP/denatonium in vitro; the paper contains no human psychophysics.

Scope: A receptor assay in a cultured-cell system (Meyerhof et al.'s results as cited by a review)

Note: EC50 is not given in the review.

In Meyerhof et al.'s cell assay, caffeine activated 5 receptors: hTAS2R7/10/14/43/46.
Limitations: Via a review. No concentration information such as EC50. The primary paper (Meyerhof 2010) is not included.

The abstract of an independent functional calcium-imaging study explicitly states, as background, "caffeine, which activates five human bitter taste receptors" (a restatement of the Meyerhof-lineage finding). In that study's own assay as well, TAS2R43/46 responded to caffeine (less sensitively than to mozambioside). This shows that the attribution of 5 receptors was still maintained as a premise in the research community as of 2020.
Limitations: The names of the 5 receptors (TAS2R7/10/14/43/46) are not listed in the abstract. The primary data are Meyerhof 2010 (the related record; full text not reached), and this row remains a restatement plus partial (TAS2R43/46) direct confirmation. The author list includes Behrens and is not independent of the Meyerhof lineage.

hop bitter acids is reported to activate human TAS2R receptor familysupported

Scope: Hop-derived bitter compounds including iso-α-acid stereoisomers, α-acids, and 8-prenylnaringenin

Note: Recorded from a structural bioinformatics paper that restates the attribution to the 3 receptors as established prior knowledge. The underlying receptor-assay source (Intelmann et al. 2009, Chemosensory Perception) could not be obtained in abstract or full text and is listed as metadata_only in sources.csv. Per-receptor entities (receptor-tas2r1 / tas2r14 / tas2r40) have deliberately not been created until the assay-level source can be read.

The abstract states that the bitterness of hop-derived compounds was found to be mediated by 3 bitter taste receptors, TAS2R1, TAS2R14, and TAS2R40, and reports a structural bioinformatics characterization of the binding modes of trans-/cis-isocohumulone, -isohumulone, -isoadhumulone, cohumulone, humulone, adhumulone, and 8-prenylnaringenin at the orthosteric binding sites of these receptors. A conserved asparagine in transmembrane helix 3 is essential for recognizing hop-derived compounds, and the surrounding binding-site residues determine ligand specificity.
Limitations: The attribution to the 3 receptors is presented as established prior knowledge rather than a new measurement in this paper, and the abstract reports no cell-based receptor assay. The underlying primary assay literature (Intelmann et al. 2009, Chemosensory Perception, DOI 10.1007/s12078-009-9049-1) could not be read in this survey and is registered as metadata_only. The docking results are computational predictions, not measured activation.

Note: The fraction is a mixture of numerous peptides; the correspondence between individual peptides and receptors cannot be identified from this experiment alone (see limits).

Cells expressing hTAS2R1, hTAS2R4, hTAS2R14, and hTAS2R16 respond to bitter fraction M (2.5 mg/mL, "tastes moderately bitter (equivalent to the bitterness of 0.125 mM of quinine)"), obtained by gel filtration of a 12-hour trypsin digest of casein (1%), with hTAS2R1 and hTAS2R4 responding especially strongly.
Limitations: The fraction is a mixture; the response is attributable only at the level of the fraction.

Limits: The authors state explicitly that the bitter fraction contains numerous structurally different peptides and is likely to include non-bitter peptides as well. In addition, background signal rises at high peptide concentrations, and hTAS2R16 showed no significant response to the synthetic dipeptides (though it did respond to its specific ligand, salicin).
Limitations: The correspondence between individual peptides and individual hTAS2Rs cannot be established with this experimental system.

Scope: Cultured primary human airway epithelium. The response is cell-autonomous within epithelial cells and does not involve taste nerves or taste perception.

Note: Detection was by microarray, RT-PCR, and anti-T2R immunofluorescence. human TAS2R receptor familyLocal intracellular calcium signals at the cell membrane. Opens large-conductance calcium-activated potassium (BKCa) channels, causing hyperpolarization of airway smooth muscle. induces bronchodilation via (contested): contested

Scope: Cultured human airway smooth muscle cells (Deshpande 2010) versus freshly isolated mouse and human native airway smooth muscle cells and tissue (Zhang 2013).

Note: Deshpande 2010 proposes local Ca2+ and BKCa activation. Zhang 2013 reports no detectable local Ca2+ events in any of 14 native cells, and that bitter compounds inhibit rather than activate BKCa channels, with 3 different BKCa channel blockers each failing to affect bronchodilation by bitter compounds; they instead propose inhibition of L-type voltage-dependent Ca2+ channels via Gβγ. The two studies differ in species and sample preparation (cultured versus freshly isolated), which itself is a candidate explanation for the discrepancy.

TAS2R-mediated relaxation is reported in association with localized [Ca2+]i response at the cell membrane opening large-conductance calcium-activated K+ (BKCa) channels, leading to hyperpolarization of the airway smooth muscle membrane.
Limitations: Cultured cells. Localized Ca2+ readout was obtained in cultured cells rather than freshly isolated cells, and conflicting research points to this as a candidate cause of the discrepancy.

Contradicts: Verbatim: "However, we subsequently demonstrated through patch-clamp recordings that bitter tastants do not activate large-conductance Ca2+-activated K+ channels but rather inhibit them. Moreover, three different large-conductance Ca2+-activated K+ channel blockers did not affect the bronchodilation induced by bitter tastants."Verbatim: "There were no detectable local Ca2+ events produced in any of the 14 cells."Verbatim: "The aforementioned two differences and another difference in which we found that bitter tastants do not activate large-conductance Ca2+-activated K+ channels strongly argue that bitter tastant-induced bronchodilation is highly unlikely to result from the generation of local Ca2+ events, which in turn activate large-conductance Ca2+-activated K+ channel."Instead, the proposed alternative is verbatim: "TAS2R stimulation activates two opposing Ca2+ signaling pathways via Gbetagamma to increase [Ca2+]i at rest while blocking activated L-type VDCCs to induce bronchodilation of contracted ASM."
Limitations: Since species and sample preparation differ from the study being refuted (freshly isolated native mouse cells versus cultured human cells), this discrepancy is not a failure to reproduce under identical conditions. This group also published a matters-arising in Nature Medicine in 2012 regarding the BK channel point, but it was not read in this investigation.

Limits: There is at least one substantial exception to the general statement that extraoral TAS2R ligands are unidentified: airway T2R38 responds to bacterial acyl-homoserine lactone-type quorum-sensing molecules produced by Pseudomonas aeruginosa and other gram-negative bacteria, which are plausible natural ligands that the paranasal sinus epithelium actually encounters, not food-derived bitter compounds.

Limitations: Because species, bitter compound, dose, and route of administration differ from rodent studies, this is not a direct refutation of mouse results but is recorded as a limitation to generalization to humans.
Source: Intragastric administration of the bitter tastant quinine lowers the glycemic response to a nutrient drink without slowing gastric emptying in healthy men (Bitarafan V; Fitzgerald PCE; Little TJ; Meyerhof W; Jones KL; Wu T; Horowitz M; Feinle-Bisset C, 2020) · abstract checked
Context: For individual agonists, only denatonium benzoate and PTC significantly increased octanoyl ghrelin, and quinine did not significantly increase total ghrelin—agonists cannot be treated as a single class effect. Verbatim: "Of the individual T2R, agonists only DB and PTC increased plasma octanoyl ghrelin levels significantly (Fig. 3C). In contrast, all T2R-agonists, except quinine, significantly increased plasma total ghrelin levels (Fig. 3D)."
Limitations: Differences in dose and pharmacokinetics per compound are not addressed within the scope of the text.

Limits: In wild-type mice, food intake after agonist administration was 49% lower than with water, but a similar decrease was observed in gustducin-knockout and ghrelin receptor-knockout mice, and the authors interpret the late-phase decrease in feeding as mediated by mechanisms independent of these pathways. Verbatim: "In WT mice, food intake was 49% lower in mice that were gavaged with T2R-agonists compared with water. A similar decrease in food intake was observed in gust-/- and GHS-R-/- mice, implying that this effect is mediated via mechanisms independent from alpha-gust"
Limitations: The authors themselves state that the framework indicated by the title does not apply to the late phase.

Show datareceptor-tas2r-family —regulates→ "Mouse gastric ghrelin secretion, short-term feeding intake, gastric emptying. Partially mediated via α-gustducin: oral administration of T2R agonists increased plasma octanoyl ghrelin in wild-type mice, but the effect was partially attenuated in gustducin-knockout mice."
