Reading the bitterness

In a heterologous-expression screen investigating mouse bitter taste receptors, 34 functionally expressed receptors were tested against mainly 128 naturally derived bitter compounds. For theobromine, a threshold concentration of 0.03 mM (mmol/L) was recorded only in the Tas2r121 column, and every other receptor column was a dash. However, no EC50 was given, and this is a value from a single study obtained with mouse receptors.

In the same study, phenanthroline activated Tas2r105, Tas2r121, Tas2r122, Tas2r135, and Tas2r144 at 1.0 mM, and in mouse behavioral tests it was also found aversive from the same concentration. Because 5 receptors responded, the aversion or overall bitterness cannot be attributed to Tas2r121 alone. Both are cell and behavioral results obtained in mice, and cannot be extrapolated to human receptors or human perception.

Checked for this overview: Matched the screening of 34 receptors and 128 compounds, the theobromine threshold of 0.03 mM, the dashes in the other columns, and the caveats of no EC50 and non-human receptors. / Matched phenanthroline at 1.0 mM, the 5 shared receptors, the behavioral threshold, and the inability to attribute this to a single receptor or extrapolate to humans.

Mechanism and receptors (2)

theobromine activates mouse Tas2r121preliminary

Scope: A heterologous-expression screen of 34 mouse Tas2r receptors against 128 mostly natural bitter compounds

Note: A non-human receptor. A single study, single value; no EC50 for theobromine has been found.

In Table 1, entitled "Threshold concentrations (mm) for Tas2r activators", the theobromine row has a single value, 0.03, in the Tas2r121 column, and every other column is a dash.
Limitations: A mouse receptor, not human. That the Table 1 columns are mouse Tas2r numbers only has been verified twice on a focused re-read. No EC50 for theobromine is given.

phenanthroline activates mouse Tas2r121preliminary

Scope: Layer: cell. A heterologous-expression screen of 34 mouse bitter taste receptors against 128 natural and synthetic bitter substances.

"Similar results were obtained for phenanthroline, which became aversive at 1.0 mm and activated all five of its cognate receptors, Tas2r105, Tas2r121, Tas2r122, Tas2r135, and Tas2r144, at a threshold of 1.0 mm." Elsewhere: "Three other mouse bitter taste receptors, Tas2r121 (11%), Tas2r135 (9%), and Tas2r144 (16%), were also responsive to a large number of different bitter compounds."
Limitations: Because this is not an agonist specific to Tas2r121 but a substance that commonly activates 5 receptors, whether the perception is mediated via Tas2r121 cannot be determined from this alone (the authors themselves state that "overall bitterness is less likely to be dependent on a single Tas2r"). A mouse receptor; cannot be extrapolated to humans.
