クラリスロマイシン

Clarithromycin is a macrolide antibiotic. Known for its strong bitterness, it is studied as a representative test subject in pharmaceutical bitterness research, particularly in studies of taste-masking coating technology for dry syrups.

Reading the bitterness

Clarithromycin is a bitter macrolide antibiotic, marketed as a dry syrup formulation (clarithromycin dry syrup, CAMD). A review of sensory evaluation methods for pharmaceuticals states that with physical coating processing, the bitterness intensity of CAMD is estimated to be reduced to approximately 1% of that of a powder suspension at the same concentration, and that approximately 99% of the bitterness was masked, as confirmed by both human sensory evaluation and taste-sensor prediction. However, the prior study cited by this review (reference 35) itself has not been independently confirmed.

Clarithromycin (CLM) microcapsules were prepared using an emulsion solvent-diffusion method with ethylcellulose as the base material, then formulated as a dry suspension. In a 3-step sensory evaluation (no bitterness / slightly bitter / bitter) conducted by 9 healthy volunteers, bitterness was reported to be sufficiently alleviated compared to the commercial CLM dry suspension. The bitterness threshold was determined at the point where half of the volunteers responded 「苦い」 (bitter) or 「やや苦い」 (slightly bitter).

pharmaceutical taste masking reduces clarithromycinpreliminary

Scope: Layer: food. A case of bitterness masking by physical coating, using the commercial clarithromycin dry syrup (CAMD) as the subject.

"Clarithromycin (CAM), known for its severe bitterness, led to the production of commercial clarithromycin dry syrup (CAMD)... estimated to be reduced to about 1% of that of an equivalent powder suspension... nearly 99% of the bitter taste was successfully masked in the dry syrup formulation, as demonstrated not only by human gustatory sensation but also by taste sensor prediction."
Limitations: The original study (reference 35) is a secondary citation from this review's description, and details of the original study's sensory panel size and methods remain unconfirmed.

pharmaceutical taste masking reduces clarithromycinpreliminary

Scope: Layer: food. Microencapsulation and dry-suspension formulation of clarithromycin (CLM) using ethylcellulose as the base material through emulsion solvent-diffusion method, with sensory evaluation conducted by 9 healthy volunteers.

"The results showed that the self-made CLM dry suspension sufficiently alleviate the bitterness of commercial CLM dry suspension (Table 4). This indicated that CLM was entrapment in the microcapsules; the bitter taste was masked efficiently in our research." "The threshold of bitterness of Self-made CLM dry suspension was determined as the point at which half of the volunteers described the taste as bitter or slightly bitter."
Limitations: Individual evaluation values by volunteer listed in Table 4 were lost during the XML conversion process; only the textual conclusion in the body text was confirmed. No statistical testing was reported.
